Design: Egg samples were obtained by collected by dissecting gravid segments of adult worms, washed several times with PBS and treated with 0,5% trypsin for 30 minutes at room temperature in order to remove any remaining contamination of adult tissues and eggs incubated overnight at 8 C. Finally, Eggs were filtered with 27 m mesh and washed extensively with PBS. Total cellular RNA was isolated and purified with TRI Reagent (Sigma Aldrich) in combination with the Direct-zol RNA Miniprep (Zymo Research), including the DNase treatment step to remove contamination with genomic DNA. RNA yield and quality were measured through an Agilent 2100 Bioanalyzer (Model G2939B). Libraries from individual samples were sequenced with Illuminas HiSeq4000 at Macrogen (Korea), by means of 100 bp paired-end reads. Libraries were prepared from total RNA, using poly(A) enrichment of mRNAs. Libraries were barcoded and multiplexed for sequencing.
